Antibacterial potency of avocado (Persea americana Mill.) seed extracts against Streptococcus viridans bacteria

Abstract

Background: Streptococcus viridans is a facultative anaerobic bacteria that can be found mostly in root canal infection. Streptococcus viridans has also been found in pulp necrosis and dental abscess. The interest of using drugs derived from herbal plants have increased, because herb lack of bacterial resistance. Persea americana Mill. seed extract has antibacterial potency, which comes from flavonoids, alkaloids, saponins, and tannins. Therefore, Persea americana Mill. seed extract can be an alternative material for irrigation in root canal treatment that could kill Streptococcus viridans bacteria. Purpose: The aim of this study was to determine the antibacterial potency of Persea americana Mill. seed extract that kill Streptococcus viridans bacteria. Methods: A serial dilution test, growth bacterial colony with streaking and spreading method was used to determine minimum bactericidal concentration (MBC) by colony counting bacteria in nutrient agar media. Growth of bacterial colonies in nutrient agar is calculated manually in colony forming unit/ml (CFU/ml). Data was analyzed statistically by one-way ANOVA and post-hoc Tukey HSD. Result: The antibacterial effect of Persea americana Mill. seed extract has reached minimum bactericidal concentration at 50% concentration, because at 50% concentration there was no bacterial growth of Streptococcus viridans. Conclusion: Persea americana Mill. seed extract has antibacterial potency with minimum bactericidal concentration (MBC) was at 50 % concentration

Abstrak

Latar belakang: Streptococcus viridans adalah bakteri fakultatif anaerob yang banyak ditemukan pada infeksi saluran akar. Streptococcus viridans juga dapat ditemukan pada nekrosis pulpa dan abses gigi. Penggunakan bahan alternatif yang berasal dari tanaman herbal kini semakin meningkat, karena rendahnya resistensi bakteri terhadap bahan herbal. Ekstrak biji Persea americana Mill. memiliki daya antibakteri yang berasal dari flavonoid, alkaloid, saponin, dan tannin. Oleh karena ekstrak biji Persea americana Mill. memiliki kandungan antibakteri tersebut, maka ekstrak biji Persea americana Mill. dapat digunakan sebagai bahan alternatif irigasi saluran akar yang memiliki fungsi dapat membunuh bakteri Streptococcus viridans. Tujuan: Tujuan penelitian ini adalah untuk mengetahui daya antibakteri ekstrak biji Persea americana Mill. dalam membunuh bakteri Streptococcus viridans. Metode: Uji daya antibakteri dilakukan dengan metode dilusi, kemudian untuk melihat pertumbuhan koloni bakteri dilakukan dengan mengamati metode streaking dan spreading yang kemudian dilanjutkan dengan penghitungan jumlah koloni bakteri pada media nutrient agar untuk menentukan besar konsentrasi bunuh minimal. Pertumbuhan koloni bakteri pada media nutrient agar dihitung secara manual dan dinyatakan dalam colony forming unit/ml (CFU/ml). Data dianalisis dengan menggunakan uji one-way ANOVA dan uji post-hoc Tukey HSD. Hasil: Konsentrasi bunuh minimal didapatkan pada ekstrak biji Persea americana Mill. 50%, karena pada konsentrasi 50% sudah tidak ditemukan adanya koloni bakteri Streptococcus viridans. Simpulan: Ekstrak biji Persea americana Mill. memiliki daya antibakteri dengan konsentrasi bunuh minimal (KBM) adalah pada konsentrasi 50%.
