Application of recombinant secretory leucocyte protease inhibitor amnion membrane on incision of Rattus norvegicus to the number of fibroblasts

Abstract

Background: Secretory leukocyte protease inhibitor (SLPI) is a protein found in various places the human body including in amnion membrane. SLPI function are to inhibit the protease, control leukocytes activity, regulate TGF-β, anti inflammatory, anti bacterial, anti retroveral (human immunodeficiency virus). The cases of incision on the oral muccosal tissue often found in Dentistry. SLPI can be used as natural biomaterial in accelerating the process of wound healing, which is characterized by an increase in the number of fibroblasts cells through the inhibiton of elastase production. Purpose: To know the application of recombinant SLPI amnion membrane fluid on incision wounds of Rattus norvegicus in increasing the number of fibroblasts on day 4. Methods: Incisions was carried out on the 24 rats (Rattus norvegicus) males, divided to 4 groups, control group (not given anything), P1 (SLPI 0.03 cc), P2 (SLPI 0.045), and P3 group (SLPI 0.06 cc). The 24 rats (Rattus norvegicus) will be executed on day 4. The samples were tested by histopathological (HPA) test to observe the number of fibroblast cells using microscope with 400x magnification. Results: The difference of fibroblasts cells number will be evaluated statistically using Kruskal-Wallis test, statistic analistic demonstrated that there are significant differences between the control and treatment groups (p < 0.05, p=0.000). Mann-Whitney statistical test demonstrated that the difference of control group and all treatment group have significant differences (p < 0.05, p = 0.04). Conclusion: Application of 0,06 cc recombinant SLPI amnion membrane on incision wound Rattus norvegicus showed the highest number of fibroblasts cells on day 4.

Abstrak

Latar belakang: Secretory leukocyte protease inhibitor (SLPI) suatu protein yang ditemukan pada berbagai tempat di dalam tubuh manusia diantaranya di dalam membran amnion. SLPI mempunyai beberapa fungsi di antaranya, menghambat protease, kontrol aktivitas leukosit, meregulasi TGF-β, anti inflamasi, anti bakteri, anti retroveral (human immunodeficiency virus). Di dalam kedokteran gigi banyak sekali ditemukan kasus luka pada jaringan mukosa rongga mulut. SLPI dapat digunakan sebagai biomaterial alami dalam mempercepat proses penyembuhan luka. yang ditandai dengan peningkatan jumlah sel fibroblast. Penelitian sebelumnya telah berhasil mengekspresikan SLPI manusia yang berasal dari membran amnion sebagai SLPI rekombinan full-length (rSLPI) di E.coli. Tujuan: mengetahui aplikasi cairan SLPI rekombinan membran amnion pada luka sayat Rattus norvegicus dalam meningkatkan jumlah fibroblas pada hari ke-4. Metode: Melakukan insisi pada 24 ekor tikus (Rattus norvegicus) jantan, kemudian membagi dalam     4 kelompok perlakuan, yaitu kelompok kontrol (tidak diberi apa-apa), kelompok P1 (SLPI 0,03 cc), kelompok P2 (SLPI 0,045), dan kelompok P3 (SLPI 0,06 cc). Pada hari ke-4, hewan coba diambil jaringannya untuk pembuatan preparat histopatologi (HPA). Kemudian dilakukan penghitungan jumlah sel fibroblas menggunakan mikroskop pada pembesaran 400x. Hasil: Berdasarkan data statistik Kruskal-Wallis, terdapat perbedaan signifikan antara kelompok kontrol dan perlakuan (p<0.05, p=0,000). Data statistik Mann-Whitney membuktikan bahwa perbedaan setiap kelompok kontrol dan semua kelompok perlakuan memiliki perbedaan signifikan (p<0.05, p=0,04) Simpulan: Penelitian ini membuktikan bahwa dengan aplikasi sebanyak 0,06 cc SLPI rekombinan membran amnion adalah aplikasi yang meningkatkan jumlah fibroblas paling tinggi pada hari ke-4.
