Cytotoxicity Test of Citrus Limon Peel Essential Oil in Human Gingival Fibroblast

Abstract

Background. Many studies have reported the effect of Citrus limon peel essential oil as antifungal and antibacterial that further can be used in dental medicine. The active components in Citrus limon peel essential oil such as D-limonene. citral, α-pinene, β-pinene, and γ-terpinene are especially known for their antifungal and antibacterial activities. However, the safety of this material has not been confirmed.  Purpose. To investigate the safest concentration that can be accepted in human gingival fibroblast cells. Method. This research was a laboratory experimental research and carried out with 7 different concentrations (essential oil with concentration of 50%, 25%, 12.5%, 6.25%, 3.12%, 1.56%, and 0.78%) and 3 different groups as control. Cytotoxicity test in this research used MTT assay method with optical density (OD) values, was read using ELISA reader, as the result. Result. Essential oil with concentration of 50%, 25%, 12.5%, 6.25%, and 3.12% was very toxic and made HGF cells could not survive at all. Whereas, essential oil with concentration of 1.56% and 0.78% was also toxic but HGF cells could survive in a very low viability. Conclusion. Citrus limon peel essential oil with concentration of 50%, 25%, 12.5%, 6.25%, 3.12%, 1.56%, and 0.78% were very toxic to be applied in HGF cells. In the future, the research of cytotoxicity with concentration under 0.78% is needed to obtain an optimal concentration that does not cause cell damaged.

Abstrak

Latar Belakang. Beberapa penelitian telah membuktikan manfaat minyak esensial kulit jeruk lemon (Citrus limon) sebagai antifungi dan antibakteri yang dapat digunakan di bidang kedokteran gigi. Kandungan D-limonene, citral, α-pinene, β-pinene, dan γ-terpinene dalam minyak esensial kulit jeruk lemon (Citrus limon) memiliki sifat antifungi dan antibakteri. Namun belum dilakukan uji sitotoksisitas bahan ini sehingga keamanan penggunaan bahan ini belum dapat dipastikan. Tujuan. Untuk mengetahui konsentrasi aman penggunaan bahan yang dapat digunakan terhadap sel human gingiva fibroblast (HGF). Metode. Penelitian yang dilakukan adalah penelitian eksperimental laboratoris dengan Post Test Only menggunakan 7 kelompok perlakuan (minyak esensial dengan konsentrasi 50%, 25%, 12.5%, 6.25%, 3.12%, 1.56%, dan 0.78%) dan 3 kelompok kontrol. Uji sitotoksisitas yang dilakukan adalah uji MTT assay dengan hasil berupa nilai Optical Density (OD) yang didapatkan dari pembacaan ELISA reader. Hasil. Didapatkan sel HGF tidak dapat bertahan hidup total pada konsentrasi 50%, 25%, 12.5%, 6.25%, dan 3.12%. Sel HGF dapat bertahan hidup dengan nilai OD sangat rendah pada konsentrasi 1.56% dan 0.78%. Kesimpulan. Minyak esensial kulit jeruk lemon dengan konsentrasi 50%, 25%, 12.5%, 6.25%, 3.12%, 1.56%, dan 0.78% masih bersifat sangat toksik terhadap sel HGF. Penelitian lebih lanjut dengan konsentrasi minyak esensial dibawah 0,78% diperlukan untuk mendapatkan konsentrasi optimal minyak esensial kulit jeruk lemon yang tidak menimbulkan toksisitas terhadap sel.
