Effect of iron exposure toward expression of spesific protein Extracellular Polymeric Substance (EPS) of Aggregatibacter actinomycetemcomitans

Abstract

Background: Aggressive periodontitis was found at high prevalence in Brazil, Iraq, Indonesia, dan United States. Trend of recent studies were to find alternative subtitutes of antibiotics which were able to inhibit or even to kill oral bacterias, but the implementation in dentistry was still less common. Purpose: Studies on bacteria biofilm would be alternative of preventive treatments in reducing the prevalence of aggressive periodontitis in society, for biofilm protects bacteria from environmental condition, such as host immune attack and antimicobrial substances. The aim of this study was to look at the effect of iron exposed towards expression of spesific protein of biofilm extracellular polymeric substance (EPS) of Aggregatibacter actinomycetemcomitans, the main bacteria which cause aggressive periodontitis. Methods: Protein containing biofilm EPS was isolated from culture of A. actinomycetemcomitans. The protein isolate underwent some procedures: electrophoresis, electroelution, rabbit immunization, serum isolation, and antibody purification. The antibody then used for Western blotting procedure in the following round of procedures. Protein containing biofilm EPS was exposed by iron, then isolated once more from culture of A. actinomycetemcomitans. The protein isolate then runned trough electrophoresis and Western blotting. Results: This study resulted in reduced expression of biofilm EPS spesific protein as a response of iron exposure. Conclusion: Iron exposure was proven to bring an impact towards expression of biofilm EPS spesific protein of Aggregatibacter actinomycetemcomitans.
