Effectiveness of Lemon Essential Oils to Prevent The Growth of Oral Microorganism in The Denture Wearer

Abstract

Background: Candida albicans and Staphylococcus aureus are microorganisms that most often found on the surface of the denture. Lemon essential oils can inhibit the growth of the Candida albicans. However, research on the effectiveness of lemon essential oil to prevent the growth of oral microorganisms in the denture wearer has not been studied. Lemon essential oil has many chemical components, such as limonene, γ-terpinene, and oxygenated monoterpinene which serves as an antimicrobes. Purpose: To know the ability of lemon essential oils in inhibiting the growth of oral microorganism’s colony found in the denture wearer. Material and Methods: Lemon essential oil was extracted from its lemon peel using cold squeezing method. Antimicrobial activity tested using a disc diffusion method. Microorganism from the oral cavity of denture wearer which is diluted as standard 0.5 McFarland is being grown on Muller Hinton media. Paper discs, each has been dipped in lemon essential oil of 70%, 80% and 90% concentrations, and sterile aquadest laid on Mueller Hinton media, then incubated for 2x24 hours. Results can be observed by measuring the diameter of inhibition zone using caliper. Result: There is significant difference between lemon essential oil in concentration 80% and 90% (p<0,05) toward oral microorganism in denture wearer. Conclusion: Lemon essential oil can inhibit the growth of oral microorganism and showed a minimum inhibitory activity of 80% (MIC = 80%).

Abstrak

Latar belakang: Candida albicans dan Staphylococcus aureus adalah mikroorganisme yang paling sering ditemukan pada permukaan gigi tiruan. Minyak esensial lemon dapat menghambat pertumbuhan Candida albican, namun penelitian tentang efektivitas minyak esensial lemon untuk mencegah pertumbuhan mikroorganisme rongga mulut pada pemakainya gigi tiruan belum diteliti. Minyak esensial lemon memiliki banyak komponen aktif, seperti limonene, γ-terpinene, dan oxygenated monoterpene yang berfungsi sebagai antmikroba. Tujuan: Untuk mengetahui efek minyak esensial lemon dalam menghambat pertumbuhan mikroorganisme rongga mulut pada pemakai gigi tiruan. Metode: Minyak esensial lemon dibuat dari kulit lemon dengan cara cold squeezing. Uji aktivitas antimikroba dilakukan dengan metode difusi. Mikroorganisme dari rongga mulut pemakai gigi tiruan yang diencerkan sesuai standar 0,5 McFarland ditumbuhkan pada media Muller Hinton. Masing-masing paper disc yang telah dicelupkan ke dalam minyak esensial lemon konsentrasi 70%, 80%, 90%, dan aquades steril diletakkan pada media Muller Hinton, kemudian diinkubasi selama 2x24 jam. Hasil dapat diamati dengan mengukur diameter zona hambat menggunakan jangka sorong. Hasil: Ada perbedaan yang signifikan antara minyak esensial lemon konsentrasi 80% dan 90% (p <0,05) terhadap mikroorganisme rongga mulut pada pemakai gigi tiruan. Kesimpulan: Minyak esensial lemon dapat menghambat pertumbuhan mikroorganisme rongga mulut dan menunjukkan konsentrasi hambat minimum yaitu 80% (MIC = 80%).
