Inhibition effect of forastero cacao bean husk extract on the growth of Streptococcus mutans

Abstract

Background. The World Health Organization (WHO) data in 2003 show that Indonesian children have high probability of having caries approximately about 60-90%. The major bacteria which is found in the process of dental caries is Streptococcus mutans (S. mutans). The growth of S. mutans should be inhibited to control caries. An alternative way to control caries is by using natural products, such as forastero cacao bean husk extract (FCBHE). The aim of this study was to determine the minimum inhibitory concentration (MIC) of FCBHE on the growth of S. mutans. Methods. This study was a laboratory experimental which used S. mutans stock. The FCBHE was extracted with ethanol and used the dilution methods to make 5 experiment groups of extract with different concentration 64 mg/ml, 80 mg/ml, 96 mg/ml, 112 mg/ml and 128 mg/ml and two control groups, positive and negative. S. mutans stock was added to different concentration of FCBHE and incubated for 24 hours at 37oC. After that, FCBHE+S. mutans was multiplied in TYC media and incubated for 24 hours at 37oC. After 24 hours, the colony of S. mutans was counted by colony counter to determine the value of MIC of FCBHE on the growth of S. mutans. There are 7 treatment groups of extract with a concentration. The data was analyzed using Oneway ANOVA test. Result. At 112 mg/ml concentration, the amount of S. mutans colony was decreased and less than 64 mg/ml, 80 mg/ml and 96 mg/ml concentration. There was none of S. mutans colony at 128 mg/ml concentration. Based on the colony counting showed that the colony of S. mutans significantly decreased as the extract concentration increased. Conclusion. Minimum concentration of FCBHE to inhibit S. mutans was 112 mg/ml.
