The effect of orthodontic mechanical force to the expressions of runx2 on tension side rat’s molar

Abstract

Background: Orthodontic tooth movement occurs by the alveolar bone remodeling in response to mechanical forces Many of the genes play a role in bone remodeling during orthodontic tooth movement, transcription factor that is important in osteoblastogenes is Runt-related transcription factor2 (Runx2), where the differentiation and maturation of osteoblasts is influenced by regulation of Runx2. Objective : To analyze the effect of orthodontic mechanical force to the expression of Runx.  Materials and method : 32 male Wistar rats were divided into control grousp and treatment groups. A closed niti coil spring was placed between the maxillary first molar and the incicors on treatment groups. An immunohistochemistry analysis were perform in each group at the following time-points: 1 day, 3 day, 6 day and 9 day. The comparison among groups was analyzed statistically using one-way analysis of variance (ANOVA). Result : Treatment group on day 1, 3,6, and 9 showed significant increased in the expression of Runx2. Conclusion : There is a difference in Runx2 expression on orthodontic tooth movement between grups.

Abstrak

Latar Belakang: Pergerakan gigi ortodonti terjadi oleh adanya remodeling tulang alveolar sebagai respon kekuatan mekanis. Banyak gen yang ikut berperan dalam remodeling tulang selama pergerakan gigi secara ortodonti, salah satunya faktor transkripsi penting dalam osteoblastogenesis seperti Runt-related transcription factor2 (Runx2), dimana diferensiasi dan maturasi osteoblas sangat dipengaruhi oleh regulasi Runx2. Bahan dan metode : 32 tikus wistar jantan di bagi menjadi 2 yaitu kelompok control dan kelompok perlakuan.Niti closed coil spring  dipasang di antara gigi insisif dan molar pertama rahang atas pada kelompok perlakuan. Masing-masing kelompok kemudian dilakukan analisa imunohistokimia pada hari ke 1, 3, 6 dan 9. Data hasil penelitian kemudian dilakukan uji beda satu arah (ANOVA) untuk menganalisis perbedaan ekspresi Runx2 dan pada masing-masing kelompok penelitian.  Hasil : Kelompok perlakuan pada hari ke-1,3,6, dan 9 menunjukkan peningkatan ekspresi Runx2 yang bermakna. Kesimpulan : Terdapat perbedaan ekspresi Runx2 antara kedua kelompok.
